Sirtuine sind mit der Pathogenese von verschiedenen Erkrankungen verbunden. In dieser Arbeit zeigten wir, dass Inhibitoren von verschiedenen Sirt-Isoformen mittels Docking und Anwendung einer kurzen MD-Simulation erhalten werden können. Eine signifikante Korrelation zwischen den geschätzten freien Bindungsenergien und den in vitro Daten wurde festgestellt. Die strukturelle Basis für die selektive Hemmung von Sirt2 wurde durch die Identifizierung eines neuen potenten Inhibitors, SirReal2, aufgeklärt. Außerdem könnte die Struktur Aktivität bzw. Selektivität Beziehung für neu entwickelten, photo-schaltbaren Sirt-Inhibitoren mittels Docking-Studien erklärt werden. Einige neue und potente smSirt2 Inhibitoren wurden auch identifiziert. Ein Homologiemodell der smSirt2 (HM) wurde generiert, und die Bindungsinteraktionen der Inhibitoren mit smSirt2-HM wurden durch Docking rationalisiert. Schließlich, etablierten wir Docking- und Pharmakophor-basierte virtuelles Screening-Methoden, die anschließend mit dem „African natural product anticancer database (AfroCancer)“ und die bekannten Targets für die Krebstherapie benutzt wurden. Sirtuins have been linked to the pathogenesis of several diseases including neurodegeneration and cancer. We demonstrated that protein-inhibitor complexes of sirtuin isoforms can be obtained by ligand docking and applying a short molecular dynamics simulation. We determined a significant correlation between the binding free energy of the compounds estimated by MM-GBSA calculations and in vitro data. Next, we established the structural basis of Sirt2-selective inhibition with a new potent drug-like inhibitor; SirReal2. Moreover, we explained the structure activity/selectivity relationship observed for the newly developed photoswitchable potent Sirt2-selective inhibitors by docking. Additionally, we identified a number of potent novel smSirt2 inhibitors. Due to the absence of the smSirt2 crystal structure, we generated a homology model (HM) for this enzyme and rationalize the binding interactions of the detected inhibitors by means of docking. Finally, we established docking- and pharmacophore-based virtual screening methods on the newly developed African natural product anticancer database and already known anticancer drug targets.
