Die vernachlässigten Tropenerkrankungen humane Afrikanische Trypanosomiasis, Chagas-Erkrankung und verschiedene Formen der Leishmaniose werden durch Parasiten der Gattungen Trypanosoma und Leishmania verursacht. Diese Einzeller besitzen einen einzigartigen Redoxmetabolismus mit Enzymen wie z. B. der genetisch und chemisch validierten Trypanothion-Synthetase (TryS). Das Ziel dieser Arbeit war es, Verbindungen basierend auf antitrypanosomalen N5-substituierten Paullonen wie MOL2008 (41) zu entwickeln, die eine verbesserte Aktivität, Selektivität und Wasserlöslichkeit aufweisen und die TryS inhibieren. Die konzipierten Verbindungen beinhalten 9-substituierte Paullon-Derivate (Serie I) und strukturell vereinfachte Benzazepinon-Derivate ohne Indol-Teilstruktur (Serie II–VIb). Enzymkinetische Untersuchungen von Medeiros et al. und molecular docking-Untersuchungen dieser Arbeit führten zu einer neuen Vorstellung eines Spermidin-kompetitiven Bindemodus TryS-inhibierender Paullone. Verbindungen der Serie I mit basischen N5-Seitenketten inhibierten die Leishmania infantum TryS (LiTryS) im submikromolaren Bereich (45a–c), tert-Butoxycarbonyl-geschützte neutrale Vorstufen hemmten die LiTryS moderat im zweistellig mikromolaren Bereich (63a–c) und Verbindungen mit einer Carbonsäure-Gruppe in der N5-Seitenkette inhibierten die LiTryS bei 30 µM kaum (62a–c). Die Viabilität von Trypanosoma brucei brucei wurde von Verbindungen der Serie I im einstellig (45a–c und 63a–b) bis niedrig zweistellig mikromolaren Bereich (62a–c und 63c) vermindert. Davon wiesen nur 62a und 63b eine Selektivität größer als zehn gegenüber murinen Makrophagen auf. Keine der getesteten Verbindungen hatte einen nennenswerten Effekt auf die T. b. brucei TryS bei 30 µM bzw. L. infantum- Amastigoten bei 20 µM. Verbindungen der Serien II–V besaßen an keinem der getesteten biologischen Systeme bedeutende Aktivität. Alle Verbindungen außer 45a und 45c erfüllten die Kriterien für hits und leads für Arzneistoffkandidaten gegen trypanosomale Erkrankungen in Bezug auf die berechneten physikochemischen Eigenschaften sowie die gemessene Wasserlöslichkeit. Die Serie II–V-Verbindungen waren besonders druglike und leichter wasserlöslich im Vergleich zu Serie I-Verbindungen. Die durchgeführten Untersuchungen haben die bekannten Struktur-Aktivitätsbeziehungen (SAR) TryS-inhibierender und antitrypanosomaler Paullone erweitert. Verbindungen ohne Indol-Teilstruktur wiesen an keinem der getesteten Systeme Bioaktivität auf. Modifikationen des 9-Substituenten am Paullon-Grundkörper führten zu Verbindungen mit erhaltener Aktivität in einer ähnlichen Größenordnung wie MOL2008 (41). Basische N5-Substituenten waren günstig für eine LiTryS-Inhibition. Mögliche zukünftige Strukturmodifikationen zur Erkenntnis detaillierterer SAR wurden vorgeschlagen. The neglected tropical diseases human African trypanosomiasis, Chagas disease and various forms of leishmaniasis are caused by parasites of the genera Trypanosoma and Leishmania. These protists possess a distinctive redox metabolism with enzymes such as the genetically and chemically validated trypanothione synthetase (TryS). The aim of this work was to develop compounds derived from antitrypanosomal N5-substituted paullones such as MOL2008 (41) that exhibit improved activity, selectivity and water solubility and inhibit TryS. The designed compounds include various 9-substituted paullone derivatives (series I) and structurally simplified benzazepinone derivatives without an indole moiety (series II–VIb). Enzyme kinetic studies by Medeiros et al. and molecular docking studies of this work led to a new concept of a spermidine-competitive binding mode of TryS-inhibition paullones. Series I compounds displayed Leishmania infantum TryS (LiTryS) inhibition, whereby basic N5-substituents led to a stronger submicromolar inhibition (45a–c). tert-Butoxycarbonyl-protected neutral precursors of the amines 45a–c inhibited LiTryS moderately in the double-digit micromolar range (63a–c) and compounds possessing a carboxylic acid in the N5-side chain hardly inhibited LiTryS at 30 µM (62a–c). The viability of Trypanosoma brucei brucei was impaired by series I compounds in the single-digit (45a–c and 63a–b) to low double-digit micromolar range (62a–c and 63c). Of these, only 62a and 63b had a selectivity greater than 10 over murine macrophages. None of the compounds tested had a significant effect on T. b. brucei TryS at 30 µM or L. infantum amastigotes at 20 µM. Compounds of series II–V did not show meaningful activity in any of the biological systems tested. All compounds except for 45a and 45c met the criteria for hits and leads for drug candidates against trypanosomal diseases in terms of calculated physicochemical properties and measured water solubility. The series II–V compounds were particularly druglike and more water-soluble compared to series I compounds. The performed studies have contributed to the expansion of the known structure-activity relationships (SAR) of TryS-inhibiting and antitrypanosomal paullones. Compounds devoid of an indole substructure showed no bioactivity in any of the tested systems. Modifications of the 9-substituent on the paullone scaffold led to compounds with comparable activity to MOL2008 (41). Basic N5-substituents were advantageous for LiTryS-Inhibition. Possible further structural modifications to gain more detailed insights into SAR were proposed.
