Breast cancer disproportionately affects women in sub-Saharan Africa, where aggressive subtypes like triple-negative breast cancer (TNBC) are significantly overrepresented. TNBC constitutes more than 30% of breast cancer cases in Benin, Gabon, and Togo, respectively, compared to 10% in France. This stark disparity suggests potential genetic factors driving TNBC prevalence among African women. However, limited genomic research in these populations hinders the development of tailored diagnostic and therapeutic strategies.FFPE samples are essential for next-generation sequencing (NGS) research, preserving tissue morphology and allowing retrospective genetic analyses, but DNA fragmentation and crosslinking present significant challenges. Our study revealed extensive DNA degradation in FFPE samples from Benin, Gabon, and Togo, with negative qPCR results or statistically higher Ct values compared to fresh biopsies (e.g., HPRT: 32.2 vs. 25.8; ALB: 33.0 vs. 24.7; TBP: 35.1 vs. 27.1; p < 0.001), limiting their suitability for high-throughput sequencing.
